Lone, M.A.; Santos, Tânia; Alecu, I.; Silva, Liana C.; Hornemann, T.Sphingolipids (SLs) are fundamental components of eukaryotic cells. 1-Deoxysphingolipids differ structurally from canonical SLs as they lack the essential C1-OH group. Consequently, 1-deoxysphingolipids cannot be converted to complex sphingolipids and are not degraded over the canonical catabolic pathways. Pathologically elevated 1-deoxySLs are involved in several disease conditions. Within this review, we will provide an up-to-date overview on the metabolic, physiological and pathophysiological aspects of this enigmatic class of "headless" sphingolipids.
- 5-(Hydroxyphenyl)-γ-Valerolactone-Sulfate, a Key Microbial Metabolite of Flavan-3-ols, Is Able to Reach the Brain: Evidence from Different in Silico, In Vitro and In Vivo Experimental ModelsPublication . However, phenolics like flavan-3-ols (F3O) are poorly absorbed along the gastrointestinal tract and structurally rearranged by gut microbiota, yielding smaller and more polar metabolites like phenyl-γ-valerolactones, phenylvaleric acids and their conjugates. The present work investigated the ability of F3O-derived metabolites to cross the blood-brain barrier (BBB), by linking five experimental models with increasing realism. First, an in silico study examined the physical-chemical characteristics of F3O metabolites to predict those most likely to cross the BBB. Some of these metabolites were then tested at physiological concentrations to cross the luminal and abluminal membranes of brain microvascular endothelial cells, cultured in vitro. Finally, three different in vivo studies in rats injected with pure 5-(3',4'-dihydroxyphenyl)-γ-valerolactone, and rats and pigs fed grapes or a F3O-rich cocoa extract, respectively, confirmed the presence of 5-(hydroxyphenyl)-γ-valerolactone-sulfate (3',4' isomer) in the brain. This work highlighted, with different experimental models, the BBB permeability of one of the main F3O-derived metabolites. It may support the neuroprotective effects of phenolic-rich foods in the frame of the "gut-brain axis".

Fortunato, Milene; Siopa, Filipa; Afonso, CarlosUsing environmentally friendly conditions, the nucleophilic ring-opening reaction of 6-azabicyclo[3.1.0]hex-3-en-2-ol with 1-methyl-1H-tetrazole-5-thiol provided a novel thiol-incorporated aminocyclopentitol, (1R,4S,5S)-5-((3-hydroxypropyl)amino)-4-((1-methyl-1H-tetrazol-5-yl)thio)cyclopent-2-en-1-ol, in excellent yield (95%). The newly synthesized compound was analyzed and characterized via 1H, 13C-NMR, HSQC, and mass spectral data.
- Theses and dissertations from ULisboa to the world : a case study on the international visibility of academic publications in the institutional repositories of the University of LisbonPublication . Melo, Luiza Baptista; Lopes, Sílvia Costa; Henriques, Susana Oliveira; Sanches, TatianaFocused on the collection of theses and dissertations, the aim of the following study is to look to the scientific production of ULisboa and analyse the international visibility of it through the statistical report of these two institutional repositories for the last 10 years (2010 - 2019). For this purpose, the total number of thesis and disserta tions of the ULisboa, archived in both repositories, were collected, for the period 2010 - 2019 (10 years of implementation of the institutional repositories). Usage statistics (views and downloads) were obtained as well as the home countries of these access es. A descriptive analysis was performed. Results showed that the number of thesis and dissertation archived in the repositories has been growing, as well as their use (views and downloads) is very significant in both repositories.
- Antimicrobial Ceramic Filters for Water Bio-DecontaminationPublication . Ferreira, Olga; Rijo, Patricia; Gomes, João; Santos, Ricardo; Monteiro, Sílvia; Guedes, Rita; Serralheiro, Maria Luísa; Gomes, Marisa; Gomes, Luciana C.; Mergulhão, Filipe J.; Silva, Elisabete R.Bio-contamination of water through biofouling, which involves the natural colonization of submerged surfaces by waterborne organisms, is a global socio-economic concern, allied to premature materials bio-corrosion and high human health risks. Most effective strategies release toxic and persistent disinfectant compounds into the aquatic medium, causing environmental problems and leading to more stringent legislation regarding their use. To minimize these side effects, a newly non-biocide-release coating strategy suitable for several polymeric matrices, namely polydimethylsiloxane and polyurethane (PU)-based coatings, was used to generate antimicrobial ceramic filters for water bio-decontamination. The best results, in terms of antimicrobial activity and biocide release, showed an expressed delay and a decrease of up to 66% in the population of methicillin-resistant Staphylococcus aureus bacteria on ceramic filters coated with polyurethane (PU)-based coatings containing grafted Econea biocide, and no evidence of biocide release after being submerged for 45 days in water. Biocidal PU-based surfaces were also less prone to Enterococcus faecalis biofilm formation under flow conditions with an average reduction of 60% after 48 h compared to a pristine PU-based surface. Biocidal coated filters show to be a potential eco-friendly alternative for minimizing the environmental risks associated with biofouling formation in water-based industrial systems.

- Codificación sistemática de contenidos educativos de farmacia prácticaPublication . Nunes da Cunha, Ines; Fernandez-Llimos, FernandoIntroducción: Los organismos internacionales farmacéuticos reclaman una educación farmacéutica más centrada en el paciente, que de apoyo a las funciones de la atención farmacéutica. Para ello, se han desarrollado marcos de competencias, que no siempre se han asociado a los correspondientes contenidos docentes. Objetivo: Identificación sistemática de los contenidos programáticos del área de farmacia práctica, mediante la creación de un árbol de codificación de contenidos. Método: Análisis cualitativo de los contenidos docentes de asignaturas del área de farmacia práctica de los planes de estudios de las facultades de farmacia de Australia, Canadá, Estados Unidos, y Nueva Zelanda. Los syllabus fueron obtenidos de las páginas web de las facultades de farmacia, excluyendo las que no tenían el plan de estudios completo disponible en inglés. Se solicitó a los responsables de las asignaturas información adicional por correo electrónico. Se creó un árbol de contenidos inicial a partir de directrices del Accreditation Council for Pharmacy Education. Se realizó un proceso iterativo de codificación dirigida, para obtener el árbol final de códigos que permitiese clasificar los contenidos docentes del área de farmacia práctica. Resultados: Se incluyeron 110 facultades (Australia=15; Canadá=5; Estados Unidos=89; y Nueva Zelanda=1), con 8733 asignaturas, de las que 1703 (19.5%) pertenecían al área de farmacia práctica y presentaban syllabus con información detallada. Se obtuvo respuesta con información adicional de 119 asignaturas. El análisis de contenido convirtió el árbol inicial de 39 categorías, en un árbol final de codificación con 3 niveles jerárquicos y 69 categorías. Conclusión: En los países del análisis, aproximadamente el 20% de las asignaturas pertenecen al área de farmacia práctica. Se ha conseguido un árbol de codificación y clasificación de los contenidos docentes de farmacia práctica. Futuros estudios deberían mapear las asignaturas actuales para determinar si los contenidos identificados están siendo enseñados.
- Readability of medicinal package leaflets: a systematic reviewPublication . Pires, Carla; Vigário, Marina; Cavaco, AfonsoTo review studies on the readability of package leaflets of medicinal products for human use. We conducted a systematic literature review between 2008 and 2013 using the keywords “Readability and Package Leaflet” and “Readability and Package Insert” in the academic search engine Biblioteca do Conhecimento Online, comprising different bibliographic resources/databases. The preferred reporting items for systematic reviews and meta-analyses criteria were applied to prepare the draft of the report. Quantitative and qualitative original studies were included. Opinion or review studies not written in English, Portuguese, Italian, French, or Spanish were excluded. We identified 202 studies, of which 180 were excluded and 22 were enrolled [two enrolling healthcare professionals, 10 enrolling other type of participants (including patients), three focused on adverse reactions, and 7 descriptive studies]. The package leaflets presented various readability problems, such as complex and difficult to understand texts, small font size, or few illustrations. The main methods to assess the readability of the package leaflet were usability tests or legibility formulae. Limitations with these methods included reduced number of participants; lack of readability formulas specifically validated for specific languages (e.g., Portuguese); and absence of an assessment on patients literacy, health knowledge, cognitive skills, levels of satisfaction, and opinions. Overall, the package leaflets presented various readability problems. In this review, some methodological limitations were identified, including the participation of a limited number of patients and healthcare professionals, the absence of prior assessments of participant literacy, humor or sense of satisfaction, or the predominance of studies not based on role-plays about the use of medicines. These limitations should be avoided in future studies and be considered when interpreting the results.
