- Different HCV Exposure Drives Specific miRNA Profile in PBMCs of HIV PatientsPublication . Valle-Millares, Daniel; Brochado-Kith, Óscar; Martín-Carbonero, Luz; Domínguez-Domínguez, Lourdes; Ryan, Pablo; De los Santos, Ignacio; De la Fuente, Sara; Castro, Juan M.; Lagarde, María; Cuevas, Guillermo; Mayoral-Muñoz, Mario; Matarranz, Mariano; Díez, Victorino; Gómez-Sanz, Alicia; Martínez-Román, Paula; Crespo-Bermejo, Celia; Palladino, Claudia; Muñoz-Muñoz, María; Jiménez-Sousa, María A.; Resino, Salvador; Briz, Verónica; Fernández-Rodríguez, Amanda; Multidisciplinary Group of Viral Coinfection HIV/HepatitisMicro RNAs (miRNAs) are essential players in HIV and HCV infections, as both viruses modulate cellular miRNAs and interact with the miRNA-mediated host response. We aim to analyze the miRNA profile of HIV patients with different exposure to HCV to explore specific signatures in the miRNA profile of PBMCs for each type of infection. We massively sequenced small RNAs of PBMCs from 117 HIV+ infected patients: 45 HIV+ patients chronically infected with HCV (HIV/HCV+), 36 HIV+ that spontaneously clarified HCV after acute infection (HIV/HCV-) and 36 HIV+ patients without previous HCV infection (HIV). Thirty-two healthy patients were used as healthy controls (HC). Differential expression analysis showed significantly differentially expressed (SDE) miRNAs in HIV/HCV+ (n = 153), HIV/HCV- (n = 169) and HIV (n = 153) patients. We found putative dysregulated pathways, such as infectious-related and PI3K signaling pathways, common in all contrasts. Specifically, putatively targeted genes involved in antifolate resistance (HIV/HV+), cancer-related pathways (HIV/HCV-) and HIF-signaling (HIV) were identified, among others. Our findings revealed that HCV strongly influences the expression profile of PBMCs from HIV patients through the disruption of its miRNome. Thus, different HCV exposure can be identified by specific miRNA signatures in PBMCs.
- Retrato e melancolia : a fisionomia da insanidadePublication . Almeida, Ana Branco de; Ramos, José Artur Vitória de SousaThe humoral theory, proposed by Hippocrates, suggests that the human being can be divided, according to their personality, in four main types - sanguine, choleric, phlegmatic and melancholic. The melancholic type, and the concept of melancholy have been widely mentioned and explored since their origin. Its prevalence, as Problem XXX dictates, in the life and mind of extraordinary men, has generated and inspired various theories, books, and works of art. In such a wide window of time, since Ancient Greece, until today, this concept inevitably suffers alterations and different interpretations of its meaning, depending on the time and culture it’s in. In this manner, all of these different facets and versions of melancholy originate different themes of this state of mind, feeling or personality. With this dissertation I wanted to reunite those different representations, focusing on portrait and physiognomy, and how melancholy translates itself in these, depending on its context, looking to identify which elements make a portrait or face melancholic. With my research I was able to, indeed, observe and recognize certain patterns and elements that, between the many meanings and nuances that melancholy acquires, are similar through many contexts and time periods
Lopes, Pedro João Vieira; Afonso, Joana Isabel Bento da SilvaThe purpose of this thesis is to analyze the artist’s creative process, more specifically in the field of comic books, as well as all the factors that influence this process, from the conception of an idea to the final product, always keeping in mind the relation between graphics and concept. Every work of art is born from an idea, which then goes through a series of transmutations, both visual and conceptual, as the artist translates it into the physical world in order to present it as clearly as possible. A historic analysis of comic books will be done – with a bigger focus on newspaper’s comic strips – as well as some references to specific works, in order to analyze the author’s own work, using the same standards
- VISTA is a diagnostic biomarker and immunotherapy target of aggressive feline mammary carcinoma subtypesPublication . Gameiro, Andreia; Nascimento, Catarina; Correia, Jorge; Ferreira, FernandoABSTRACT - Feline mammary carcinoma (FMC) is a common neoplasia, showing aggressive clinicopathological features, without viable therapeutic options. The study of tumor microenvironment has gained importance, due to the ability to control tumor progression by regulating the immune response. Considering the lack of knowledge, feline serum VISTA levels from cats with mammary carcinoma were compared with healthy controls, and with serum levels of PD-1/PD-L1, CTLA-4, LAG-3, IL-6, and TNF-α. In parallel, VISTA tumor expression was evaluated in FMC samples. The obtained data revealed that serum VISTA levels were significantly higher in cats presenting HER2-positive (p = 0.0025) or triple-negative subtypes (p = 0.0019), with higher serum levels in luminal A (p = 0.0025) correlated to the presence of metastasis (p = 0.0471). Furthermore, in HER2- positive or triple-negative tumors, correlations were obtained between serum VISTA levels and the serum levels of the above-mentioned molecules. In tumors, VISTA expression revealed a stronger intensity in cancer cells, when compared to TILs (p < 0.0001). Stratifying the samples by subtypes, a higher number of VISTA-positive TILs was observed in the HER2-positive subtype, compared with triple-negative tumors (p = 0.0138). In conclusion, results support the development of therapeutic strategies for HER2-positive and triple-negative FMC subtypes, reinforcing the use of cats as a human oncology model.
- Discussão sobre as dificuldades tradutológicas do livro The Crack of Doom de Robert CromiePublication .
