Nanotechnology is considered an important area of interest in delivery systems. During the last decades, lipid vesicles have been deeply investigated attending that they are one of the most successful drug carrier systems. Their potential to overcome the low skin permeability enables them to provide an efficient dermal and transdermal delivery of therapeutically active hydrophobic or hydrophilic drugs. This review highlights the wide therapeutic applications of lipid vesicles focusing their evolution since the first generation as well as preparation and characterization methods (including topical delivery studies), toxicological and regulatory aspects. Lipid vesicles offer several advantages especially the improvement of therapeutic index, and consequently, the quality of life for several patients. Actually, they have been increasingly used as a strategy to reduce the side effects of skin cancer treatment. The future task will pass through the scale-up production to supply the market quality and security demands. Notwithstanding, a compromise will be needed from the regulatory agencies to upgrade their legislation about nanotechnology.
