As doenças neurodegenerativas são condições debilitantes e em grande parte intratáveis, muito pela diminuição da capacidade de autorreparação celular. Vários fatores genéticos e ambientais são responsáveis pela patogénese destas enfermidades. Apesar das diferenças nas manifestações clínicas, o processo a nível celular apresenta pontos comuns: início predominantemente durante a meia-idade ou idade avançada, influência hereditária ou esporádica e agregação e deposição intra ou extracelular de proteínas alteradas. Esta revisão teve por objetivo principal averiguar a contribuição da excitotoxicidade, particularmente, na Esclerose Múltipla e na Esclerose Lateral Amiotrófica e por que mecanismos.
Neurodegenerative diseases are debilitating and almost untreatable conditions, mostly due to the decrease in the cells' self-healing capacity. Several genetic and environmental factors are responsible for the pathogenesis of these diseases. Despite the differences in clinical symptoms, the process presents common points at the cellular level: starting predominantly during middle or old ages, of hereditary or sporadic influence, aggregation and deposition of both intra and extracellular altered proteins. The main focus of this study was to investigate the contribution of excitotoxicity, particularly in Multiple Sclerosis and Amyotrophic Lateral Sclerosis and through which mechanisms.
