As infeções fúngicas são uma das principais preocupações atuais em termos de saúde global,
com taxas de incidência e mortalidade em crescimento, especialmente em indivíduos
imunocomprometidos. Apesar do seu impacto substancial nos cuidados de saúde globais, o
panorama atual dos fungos e em contrapartida, do tratamento antifúngico, é frequentemente
subestimado, uma vez que a investigação global tende a deslocar-se para outras áreas da
microbiologia. Além disso, a atual crise de resistência antifúngica em fungos patogénicos
conhecidos, ameaça agravar ainda mais esta situação problemática, tornando potencialmente
ineficazes as limitadas classes de antifúngicos disponíveis.
As atuais classes de antifúngicos, por si só, também apresentam um espetro de atividade
complexo e efeitos secundários adversos notáveis que podem limitar a sua administração
A presente monografia apresenta uma análise global do antibiótico, Nistatina, particularmente
no seu desenvolvimento histórico, mecanismo de ação, espetro de atividade e aplicações
clínicas.
O objetivo desta monografia consiste em investigar de que forma a Nistatina pode ser
incorporada em novos sistemas de administração ou como a sua síntese pode ser ajustada,
originando novos derivados “Nystatin-like” eficazes, tornando-a pioneira no futuro das
estratégias antifúngicas no combate às infeções.
O presente trabalho conclui com uma reflexão sobre o estado dos atuais tratamentos
antifúngicos e como, ao combinar sistemas inovadores de administração de fármacos com
novas formulações, a Nistatina poderá tornar-se um num tratamento antifúngico eficaz através
de uma formulação segura e possivelmente apta para administração parentérica.
Fungal infections are a current major global health concern with increasing incidence and mortality rates, especially in immunocompromised individuals. Despite their substantial impact on global healthcare, the current landscape of fungi and, conversely, antifungal treatment, is often underestimated, as global research tends to shift to other areas of microbiology. Furthermore, the ongoing emergence of antifungal resistance in known pathogenic fungi threatens to further exacerbate this problematic situation by potentially rendering the already limited antifungal treatment classes ineffective. The current classes of antifungal drugs themselves also have a limited spectrum of activity and notable adverse side effects that may limit their administration This literature review presents a global analysis of the antibiotic, Nystatin, particularly in its historical development, mechanism of action, spectrum of activity and clinical applications. The goal of this research is to investigate how Nystatin can be incorporated into novel drug delivery systems or its synthesis tweaked to create new effective Nystatin-like derivatives. Showcasing that a safe, effective and possibly parenteral formulation is possible. The present work then concludes by reflecting on the status of antifungal treatment and the prospects for Nystatin in pioneering the future of antifungal strategies in combating infections. By combining innovative drug delivery systems with novel formulations, Nystatin could become a highly effective and potentially safe antifungal treatment for the future.
