As doenças oculares externas causadas por infeções bacterianas secundárias são comuns nos
cães. Devido às características próprias dos olhos, ao pequeno volume lacrimal e à contínua
drenagem e eliminação da mesma, o tratamento passa muitas vezes pela aplicação frequente
de colírios. Cada vez mais se desenvolvem métodos que permitem uma libertação contínua de
fármacos de forma a aumentar a adesão dos donos dos animais à terapêutica e assim, não
comprometer os tratamentos. Neste trabalho foram avaliadas as características físicas, os
índices de absorção e de libertação dos discos de quitosano incorporados com três antibióticos
(Levofloxacina, Gentamicina e Ceftriaxona) em diferentes concentrações. Verificou-se a
existência de heterogeneidade entre discos e que o índice de absorção era relativamente
grande quando comparado ao de outros estudos, facto possivelmente decorrente da origem
dos mesmos. A libertação segundo o método de TSA permitiu verificar que as zonas de
inibição durante 24 horas foram superiores ao intervalo de referência para os discos de
Levofloxacina e Gentamicina. Devido a limitações técnicas, a quantidade de discos analisada
foi pequena e nem todos os discos foram avaliados nos diferentes testes. São assim
necessários mais estudos e aperfeiçoamento dos pensos oculares antes da sua utilização na
clínica de pequenos animais.
ABSTRACT - PRELIMINARY DEVELOPMENT AND EVALUATION OF A SYSTEM FOR RELEASE OF ANTIBIOTICS BASED ON CHITOSAN FOR USE IN SUPERFICIAL OCULAR INFECTIONS - The external ocular diseases caused by secondary bacterial infections are common in dogs. Due to the characteristics of the eyes, like the residual tear volume and continuous drainage, the treatment often depends on the frequent use of eye drops. The evaluation studies on the methods for continuous drug release in order to enhance the compliance of the pet owners to not compromise the treatments have become more frequent. In this study were analysed, in terms of physical characteristics, rate of absorption and release chitosan inserts incorporated with three antibiotics (Levofloxacin, gentamicin and ceftriaxone) at different concentrations. In this work it was found that there was heterogeneity among discs and the absorption rate was relatively increased when compared to other studies, a fact due probably to its origin. The antibiotic release according to the TSA method has shown that the inhibition zone was larger than the reference interval for the levofloxacin and gentamicin discs during 24 hours. Technical limitations diminished the amount of discs analysed and not all discs were evaluated in the different tests. More studies and improvement of the ocular inserts would be needed before its clinical use.
