NKCC1 and KCC2 CO transporters in prenatal exposure to alcohol : an immunohistofluorescence study

Resumo(s)

It is known for a long time that prenatal exposure to alcohol alters the plasticity of hippocampus and leads to fetal alcohol syndrome (FAS), a condition that is characterized for growth retardation and CNS dysfunction. Previous studies in GRAP laboratory demonstrate that is possible that prenatal exposed to alcohol rats have a problem with the maturation of the GABAergic signal. It is also known that GABAergic maturation depends on the development of two chloride co transporters: NKCC1 and KCC2. This investigation attempts to establish a protocol to allow the study of the level of expression and localization in the CA1 area of hippocampus of the co transporters. The experiments performed allowed to write an immunohistofluorescence protocol that uses specific antibodies for NKCC1 and KCC2, as well as for GABAergic neurons and glutamatergic neurons, allowing the co localization of the co transporters with different types of neurons. The software used to capture pictures allows the study of the level of expression of the co transporters through the intensity of the labeling.
