Técnicas de estudo micromorfológico e molecular aplicadas ao género Hypericum

Resumo(s)

The aim of this work was the micromorphological and molecular characterization of some species of Hypericum. The morphological studies were accomplished on: H. calycinum, H. canariense, H. grandifolium, H. x inodorum and H. patulum. The molecular analysis was extended to two more species: H. androsaemum and H. perforatum ssp. perforatum. The morphological analysis were carried out by optical and scanning electron microscopy , and histochemical studies were done with reagents for the main groups of primary and secondary compounds. The epidermal characters such as size of epidermal cells, size and shape of stomata, stomatic index and presence of secretory cells are similar in all taxa. There were differences in the shape of the walls of epidermal cells, density of the cuticular waxes, index of glands and size of the mesophyll. The detected histochemical compounds are similar, except in the taxon H. patulum, which has steroids in glands (with antimony trichloride). In the study we used molecular markers ISSR and proceeded to the analysis of the sequences of the ITS region. The ISSR marker might have revealed a high degree of polymorphism and showed that the species H. inodorum and H. grandifolium are very close and the rate H. patulum and H. calycinum are species that are more remote, both morphological and molecular.
