Evaluation of Listeria monocytogenes biofilm formation: comparison between persistent and sporadic strains

dc.description.abstractL. monocytogenes has proven to be persistent in several food industries, with different environmental conditions, including nutritional availability, raising the important question: which is the major factor associated with persistence and can this be related to disinfectant susceptibility. A group of 10 persistent and 10 non persistent strains from different origins were selected and tested for biofilm forming ability using microtiter plate assay crystal violet (CV) and stainless steel coupon (SSC) methods at 25 ºC and 11 ºC for 48 h and 7 days, respectively. Susceptibility of a subset of 12 of these strains was tested by log reduction of cell enumeration from stainless steel coupons exposed to two commercial hydrogen peroxide acidic based disinfectants (P3 OXONIA and MIDIA SAN 315). Results showed no tendency between persistence traits and biofilm forming ability, even though significant differences (p<0.05) were found between both incubation temperatures. Simulation of food industry conditions (cold and nutritional stress) with biofilms on SSC also showed no relation between persistence traits and high biofilm forming ability. Susceptibility results corroborate these results by showing no significant differences in disinfectant susceptibility between persistent and non persistent strains, although P3 OXONIA with acetic and peracetic acid proved to be more effective than MIDIA SAN 315 with citric acidpt_PT
