- Contribution to the study of prognostic factors in canine oral squamous cell carcinomaPublication . Mestrinho, Lisa Alexandra Pereira; Niza, Maria Manuela Grave Rodeia Espada; Peleteiro, Maria da Conceição da Cunha e VasconcelosThe main goal of this clinical work was to contribute to the study of oral squamous cell carcinoma (OSCC) in the dog with regards to molecular and differentiation events, and their relationship with the tumor’s biological behavior – impact on the staging, prognosis or treatment. From the assessment of histological differentiation and proliferation index in a 36 OSCC case series, it was possible to identify a significant association between the proliferating cell nuclear antigen (PCNA) and Ki-67, distribution pattern and histological differentiation. PCNA expression was significantly higher than Ki-67 and a higher proliferation index was associated with poorly differentiated OSCC, resembling what is observed in humans. With regards to intra-oral locations, poorly differentiated tumors were significantly associated with the tonsillar location. Proliferation index was not associated with any of the intra-oral locations evaluated, but it was found that metastatic lymph nodes were significantly associated with OSCC with higher poliferation index. OSCC prognosis was assessed in a 13 case series where the age of the animal and tumor size, location, grade and proliferation were evaluated as post surgical prognostic factors. All cases were standardized – CT staged and clinically managed over a period of 2 years, with bone invasion, no lymphatic invasion nor distant metastasis. The results obtained indicated that grade and proliferation index were related with post operatory prognosis. Animals with high-grade tumors had shorter disease free survival times when compared to intermediate grade tumors. Also animals with high-grade tumors and with high PCNA proliferation index showed a significantly worse post-surgical prognosis. The immunohistochemical expression of the cell adhesion molecule E-cadherin and p63 protein were assessed and related with histological and clinical features at time of diagnosis in a 22 case series. Changes in p63 and E-cadherin expression seem to be frequent molecular events in canine OSCC. It was observed that p63 was overexpressed in all cases and E-cadherin was found to be underexpressed in most cases. Furthermore, p63 expression was related to the proliferation and there were different expression patterns related to tumor grade. None of the histological or immunohistochemical parameters were associated with tumor size, bone invasion or lymph node metastasis. More than the contribution to the knowledge of some of the molecular events involved in this tumor, this work explores the clinical applicability of such markers, especially proliferation, in a practical veterinary clinical setting, ilustrated in this work with one case report. This thesis also explores the potential use of the dog as a model in comparative oncology studies.
- Caracterização material dos suportes e adesivos de painéis de azulejo do Museu Nacional do Azulejo para a sua intervenção de conservação e restauroPublication . Mortari, Camila, 1990-; Manso, Marta; Esteves, LurdesThis internship project’s main theme is centered around contemporary Portuguese Glazed Tilework and the different approaches to the handling and display of its panels when relocated from their point of origin to a museological setting. It describes the process of restauration of three glazed tile panels one authored by João Segurado, another by Almada Negreiros and a small panel comprised of ceramic tiles from the early 16th century. This study focused itself toward the understanding of problems related to the materials or support structures and how they interact with the tiles, more specifically regarding deterioration. Through this investigation it became clear that a further study of these support structures’ materials was needed, so as to not compromise the integrity of the tiles. This was achieved through a combination of both the use of scientific analysis and historic heritage research. As a result, at the end of this internship, it was possible to grasp the complexity of the work the professionals go through at the laboratory of restauration and conservation of art at MNAz. Furthermore it allowed for a better understanding of the finer details of displaying works of art to the public, while both maintaining its educational facet and promoting its aesthetic experience. The knowledge acquired in this period made this internship a unique learning opportunity that provided invaluable professional growth
- Evaluation of the cost-benefit of the implementation of PAT systems for the production of solid forms in a pharmaceutical companyPublication . Cordeiro, Pedro Miguel António; Lopes, João Almeida; Bica, AntónioProcess Analytical Technology (PAT) is defined by the FDA in the PAT initiative issued in 2004 as ”a system for designing, analyzing, and controlling manufacturing through timelymeasurements (i.e.,during processing) of critical quality and performance attributes of raw and inprocess materials and processes, with the goal of ensuring the final product quality”. Since then, the pharmaceutical companies have been adopting gradually this technology in their manufacturing lines. PAT tools are indeed well recognized as a critical tool for achieving better process and product control leading to the possibility of real time release testing. This thesis evaluated the technical and economic feasibility of implementation of PAT systems into a Portuguese pharmaceutical company manufacturing line of solid forms. To accomplish this task, two products were used as models. These products were selected since they cover most unit operations of the solid forms installed manufacturing line. The work was divided in two parts. The first part is concerned with the identification of critical quality attributes (CQAs) and critical process parameters (CPPs) of the two model products in order for the proposition of opportunities of implementation of PAT strategies. These opportunities are described in detail and evaluated according to a series of criteria. Nine opportunities were identified, mainly involving NIR and RAMAN spectroscopy as suitable technology. The implementation of the identified opportunities, is expected to permit the possibility of real time release for the two products. The second part focuses on the economical feasibility of the identified PAT implementation. The economic analysis indicated that the implementation of all opportunities will reduce manufacturing / quality control time and costs substantially. The expectation of return-of-investmentis achieved in a time frame between 5 to 8 years respectively considering optimistic and pessimist scenarios.
- Jorge Barradas e seus caprichos : conservação e restauro de um painelPublication . Oliveira, Adriana Anselmo de, 1976-; Manso, Marta Cunha Monteiro, 1978-; Esteves, LurdesJorge Barradas (1894-1971) was an artist and ceramist, from the first generation of Por-tuguese modernists, well known from 1940 to 1970 for his innovations in modern wall glazed tiles. The present report aims at registering the interventions of conservation and restoration on a tile panel by this ceramist. The panel, which belongs to the collection of the Na-tional Tile Museum in Lisbon, is dated from 1963 and consists of a wall panel of glazed tiles with a decorative floral composition, one of a number of works referred to as Ca-prichos by the artist. The tiles, very fragmented, were subjected to analysis using energy dispersive X ray fluorescence spectroscopy (EDXRF), scanning electron microscopy with energy disper-sive spectroscopy (SEM-EDS) and Raman spectroscopy for material identification of the polychrome tile pieces. In this project, the historical research on the artist's work, the practice of intervention and the technical research were carried out simultaneously. The objectives were to un-derstand the panel's origin and its integration throughout the artist's whole work, the main characteristics of the materials used in the tiles, and, at the same time, preparing a diagnosis leading to the intervention and permitting future exhibition
