Lisboa: CBAA/ISA, 2009http://hdl.handle.net/10400.5/3526Relatório da Bolsa de integração na Investigação (BII)In root tip cells from seedlings of Secale cereale exposed to high temperatures (heat stress), marked modifications in nuclear morphology and microtubule cytoskeleton dynamics are observed. Seedlings were developed in normal environmental conditions at 21ºC, and transferred for 40ºC during four hours for the heat shock treatment. Root-tips were excised just after the treatment, and after distinct periods of recovery (RT). The detailed characterization of meristematic cells nuclear morphology revealed striking modifications some of which are maintained even after 24 of RT. These alterations seem to be associated with changes in cell cycle. Imunocytochemistry was used to pursue the study of microtubules (MTs) dynamics in heat treated plants using alfa-tubulin antibodies. These revealed great alterations in MTs organization, preferentially affecting interphase cells. These results clearly indicate for the first time a direct impact of heat stress on nuclear dynamics and cell cycle progressionporSecale cerealeheat stresschromatincytoskeletonAnálise do efeito das altas temperaturas na organização da cromatina e do citoesqueleto microtubular em células da raiz de Secale cerealereport
