This enzyme is responsible for leukotrien production, through the arachidonic acid pathway, and its role on physiopathological processes has been extensively reviewed. The experimental techniques for its detection and quantification, were described and their advantages and disadvantages were accounted for. They were further divided in continuous methods, where changes in the reaction mixture are monitored, and fixed time methods, which involve sample to be withdrawn from the reaction mixture. Throughout all the collection data, we could conclude that the best and most common, method for 5-LOX activity monitoring was the spectrophotometric method with diene detection at 234 nm. However, there were many differing characteristics to this experimental protocol, the absence of an enzymatic kinetic analysis and the lack of comparability between lipoxygenase from a vegetable source and 5-LOX recombinant human. Therefore, as a final result a detailed and simple adapted methodology, specially developed for the human enzyme, is purposed by us.
