Os distúrbios do sono estão entre os problemas de saúde pública mais comuns, sendo responsáveis por uma diminuição significativa da qualidade de vida dos indivíduos afetados. Existe uma enorme diversidade de distúrbios do sono, sendo a insónia o mais frequente, e caracterizado por dificuldade em dormir ou total ausência de sono. Por outro lado, a narcolepsia, caracterizada por um excesso de sono patológico, é uma patologia rara, podendo tornar-se altamente incapacitante. A terapêutica farmacológica atual para ambas as patologias tem-se revelado eficaz; porém, os seus efeitos adversos podem, por vezes, ser muito graves. Desta forma, o desenvolvimento de fármacos com um melhor perfil de segurança tem-se tornado uma preocupação crescente. Deste modo, têm vindo a ser investigados e desenvolvidos novos fármacos, capazes de atuar de forma mais seletiva, aliados a um conhecimento mais profundo da fisiologia associada aos ciclos de sono e vigília.
Sleep disorders are one of the most common public health problems, which are responsible for decreasing greatly the quality of life of the affected ones. There is a huge diversity of sleep disorders, being insomnia the most frequent among them and characterized by inability to sleep or a total lack of sleep. On the other hand, narcolepsy, which is characterized by a pathologic excess of sleep, is a rare disease and might become highly disabling. The current pharmacological treatments, for both disorders, seem to be effective; however, their side effects can, sometimes, be very severe. Thus, the development of drugs with a better safety profile has become a growing concern. Therefore, new drugs have been researched and developed, to aim more selective targets, allied to a more deepened knowledge of the sleep and wakefulness cycle physiology.
