A sua acção depende da via de sinalização por Fgf. Os estudos realizados neste trabalho visam perceber a influência, a nível do desenvolvimento do membro em vertebrados, de elementos de uma família de proteínas denominada Flrt. Esta família é composta por 3 elementos, Flrt1, Flrt2 e Flrt3 e está ainda pouco estudada. São proteínas transmembranares caracterizadas por motivos de repetições ricas em leucinas na sua parte extracelular. Existe ainda um domínio intracelular, pouco estudado, mas que se sabe ser essencial ao papel desta família como regulador da actividade da sinalização por Fgf. Flrt3 é o membro mais bem caracterizado. Em Xenopus laevis foi verificado que Flrt3 é regulado positivamente por Fgf, e sendo ele próprio um regulador positivo desta via. Em galinha Flrt3 é essencial à correcta expansão do eixo proximal-distal do membro em desenvolvimento, sendo regulado por Wnt. Este gene é expresso na AER em galinha e ratinho. Em Peixe-zebra a sua função ainda não é completamente percebida. Este trabalho de investigação consiste na clonagem de dois membros da família Flrt (Flrt2 e flrt3) e análise da expressão destes genes em embriões de Peixe-zebra nos estádios 32 e 48 horas pós fertilização. Os resultados não mostraram marcação para o gene Flrt2 e para Flrt3 sugerem uma marcação no mesênquima, ao contrário do que ocorre com outros modelos. The studies conducted within this research project aim for the understanding of the influence exerted by elements from the Flrt protein family on the limb development in vertebrates. This family consists of 3 elements - Flrt1, Flrt2 and Flrt3 - and still needs further study. These are transmembrane proteins identifiable by their extracellular leucine-rich repeat motifs. There is also an intracellular domain, yet to be further studied, that is known to be of vital importance to the part played by this family as a regulator of Fgf signalling activity. Flrt3 is the best characterised member of this family. In Xenopus laevis, Flrt3 was proved to be positively regulated by Fgf, and acts itself a positive regulator of this signaling pathway. In chicken embryos Flrt3 is vital to the correct expansion of the proximal-distal axis from the developing limb, being regulated by Wnt. This gene shows expression in AER from chicken and mouse. In zebrafish its function is not yet fully understood. This project consists in cloning two members of the Flrt family (Flrt2 and flrt3) and analysing the expression pattern of those genes in zebrafish embryos at early stages -32 and 48 hours after fertilization. The results have not shown any signal for the Flrt2 gene and suggest a signal in the mesenchyme for Flrt3, as opposed to what occurs with the other models.
